BindWorks™ is an interactive computer program for analysis of Binding Isotherms using Isothermal Titration Calorimeters (ITC). BindWorks runs under Microsoft Windows and offers the following features:

- Bindworks data file format (*.bnw) can read files created by Microcal Omega software. The program can also read text format data files originating in software spreadsheets (Microsoft Excel, Lotus 1-2-3, Quattro Pro, etc) and mathematical programs (Mathcad).
Modeling
Each model contains a set of variable parameters. Curve fitting produces optimum values for these parameters. These values will best represent the given input data set.
In addition to user-defined binding models, BindWorks supports three intrinsic binding models:
- Independent Set of Multiple Binding Sites
- Two Sets of Multiple Binding Sites
- Cooperative Model
Independent Set of Multiple Binding Sites
The Independent Set of Multiple Binding Sites is the most common model for a binding experiment. The analytical solution for the total heat is determined by the formula:

This model assumes the macromolecule has many independent binding sites (at least two). Each site can be one of the two types. Each type has its characteristics (binding constant and enthalpy). The reaction pathways are shown on the graph below.
Cooperative Binding Model
This model assumes at least two binding sites on the macromolecule as well as interaction between the two ligand molecules.
Input Data Editor
BindWorks contains an embedded spreadsheet and calculator to edit your data set. The spreadsheet contains five columns. The number of rows is equal to the number of injections in the input set. Each row corresponds to one injection.
User Defined Binding Models
BindWorks supports external, user-supplied models. During the install procedure BindWorks installs all required support files. These files, the associated documentation, and average programming experience in both Windows and C programming language environments allow users to create additional models.
Planning the ITC Experiment
Our experience shows time spent planning an experiment can exceed the time spent on data analysis. To alleviate this problem, BindWorks includes the Experiment Design Wizard. The Experiment Design Wizard is a tool that provides ITC experiment simulation based on specified ranges of expectancies for a selected binding model.
